Unique left Holy Loch after refitting and set out to patrol the Bay of Biscay in October 1942. She parted from her escort off the Scillies on 9 October and was never seen again. A sister submarine in the area heard underwater explosions and suspected an attack. Neither a German sinking claim nor a corresponding British air report establishes what happened. Unique was reported overdue on 24 October after failing to arrive at Gibraltar, ending a career that had included Italian merchant sinkings and an unsuccessful attack on Littorio.
